  The Octet system by ForteBio uses Bio-Layer Interferometry (BLI), a label-free technology for measuring biomolecular interactions. It is an optical analytical technique that analyzes the interference pattern of white light reflected from two surfaces: a layer of immobilized protein on the biosensor tip, and an internal reference layer. Any change in the number of molecules bound to the biosensor tip causes a shift in the interference pattern that can be measured in real-time.

 

Epitope Mapping using the Octet

 

The Octet is a fast and easy way to identify antibodies that bind to overlapping or spacially distinct epitopes.  Antibody 1 on the sensor captures the antigen.  Antibody 2 is then either blocked from binding by Antibody 1, or is free to bind to a different epitope. 

 

In the following example, the first antibody GMA 8019 binds to sensors A12-D12. Next, FVIII antigen is captured by GMA-8019. After blocking with control IgG each sensor is incubated with a different second antibody. Antibodies GMA-8017 and GMA-8018 show binding (sensors B12 and C12) while GMA-8010 (sensor A12) and GMA-8019 (sensor D12) don't bind to captured FVIII.
